Start with the basics. You need fuel, air and and ingnition source for combustion to occure. Air is pretty much a givin. Start with the spark, you can pull a plug wire off a sparkplug and shove a small bold inside to make contact with the connector. Hold the wire (by the rubber part not the bolt ) within a half inch or so of the engine block or other grounded metal and have someone crank the entine. If its fireing a spark you've got that part so move on to the fuel. It's harder to check on a fuel injected vehicle but you can remove the filter like you mentioned and see if its pumping fuel.

Ditto, I always do a basic tune up on a new to me vehicle. Usually like $75-$100 for a jeep. Worth it's weight in Gold!!!
